Vitamin D3 and K2 together — is it always a good combination?

Sources: 13 · Article: 18 518 characters

Jan Jaśkowski recommends combining vitamin D3 with K2, including in the form of MK7, for their combined effects and support for bones.

  • According to him, D3 increases calcium absorption, while K2 helps direct it to the right places and protect against its buildup in the arteries.
  • Jan Jaśkowski recommends 20 µg of K2 for every 1000 IU of D3 and advises against taking extra calcium.
  • Jerzy Jaśkowski gives a dose of about 500 µg of K2 per day alongside the described dose of D3.

D3 increases calcium absorption, while K2 is said to direct it to the right places and protect against its buildup in the arteries.

„Vitamin D3 increases calcium absorption, but K2 directs it to the right places. This means that the combination of D3 and K2 not only supports normal mineralization of the skeletal system, but also protects against calcium buildup in the arteries. For this reason, many experts recommend taking both vitamins at the same time, especially during long-term supplementation with high doses of vitamin D3.”Witamina D3 zwiększa wchłanianie wapnia, ale to K2 kieruje go we właściwe miejsca.

Jan Jaśkowski recommends 20 µg of K2 for every 1000 IU of D3.

„For this reason, many experts recommend taking both vitamins at the same time, especially during long-term supplementation with high doses of vitamin D3. A practical rule is to take 20 µg of vitamin K2 for every 1000 IU of vitamin D3. These proportions support proper calcium balance in the body and maximize the effects of supplementation.”Z tego powodu wielu ekspertów zaleca przyjmowanie obu witamin równocześnie, zwłaszcza przy długotrwałej suplementacji wysokimi dawkami witaminy D3. Praktyczna zasada brzmi: na każde 1000 IU witaminy D3 należy przyjmować 20 µg witaminy K2. Takie proporcje wspierają prawidłową równowagę wapnia w organizmie i maksymalizują efekty suplementacji.

Jan Jaśkowski recommends taking D3 and K2 together because of their synergistic effects and support for bones.

„Vitamins D3 and K2 MK7 have synergistic effects in the body, so it is worth taking them together. To maintain healthy bones (D3 and K2) and normal muscle and immune system function (D3). Tablet form — capsules and drops are also available.”Witaminy D3 i K2 MK7 mają synergiczne działanie dla organizmu — dlatego warto przyjmować je razem. Dla utrzymania zdrowych kości (D3 i K2), prawidłowego funkcjonowania mięśni i układu odpornościowego (D3). Wersja w tabletkach — dostępne również kapsułki i krople.

Jan Jaśkowski recommends taking D3 with K2 to avoid calcification of soft tissues and advises against taking extra calcium.

„Remember to supplement vitamin D with vitamin K2 to avoid calcification of soft tissues, including blood vessels. For this reason, you should NOT take additional calcium supplements.”Należy pamiętać aby witaminę D suplementować z witaminą K2, aby uniknąć zwapnienia tkanek miękkich w tym naczyń krwionośnych. Z tego też powodu NIE należy suplementować dodatkowo wapnia.
Jan Jaśkowski

Jan Jaśkowski recommends K2 MK7 together with D3, including when taking high doses of D3.

„So instead of being afraid of vitamin D3 toxicity, we should focus on reaching an optimal level and maintaining it throughout the year. How can you do this? We recommend products from Mojej Apteczki — containing a high concentration of vitamin D3 from lanolin along with vitamin K2 MK7, which should be taken together with vitamin D3.”Dlatego zamiast bać się toksyczności witaminy D3, powinniśmy skupić się na tym, aby osiągnąć jej optymalne stężenie i utrzymywać je przez cały rok. Jak to zrobić? My polecamy produkty Mojej Apteczki — zawierające wysokie stężenie Witaminy D3 z lanoliny wraz z witaminą K2 MK7, którą należy suplementować wspólnie z witaminą D3.
Jerzy Jaśkowski

Jerzy Jaśkowski recommends taking K2 at the same time as the described dose of D3 and gives a dose of about 500 µg of K2 per day.

„Another scientific study shows that administering a dose of 50 000 IU for 6 months resulted in a remission lasting almost 6 years. Of course, you should remember to take vitamin K-2 at the same time, in an amount of about 500 mcg per day. You should also include salads made with cauliflower, broccoli, and Brussels sprouts with peas, etc. in your diet.”Inna praca naukowa dowodzi, że podawanie dawki 50 000 UI przez 6 miesięcy, spowodowało praktycznie 6-letnią remisję. Oczywiście nie należy zapominać o przyjmowaniu jednocześnie witaminy K-2 w ilości ok. 500 mcg dziennie. Należy także, do diety włączyć surówki z kalafiora, brokułów i brukselki z groszkiem itd.

In postmenopausal women with osteoporosis, the combination of D3 and K2 increased spinal bone density more than either vitamin alone or calcium.

Aim of the study: The effect of the combined administration of vitamin D3 and vitamin K2 on bone mineral density (BMD) of the lumbar spine was examined in postmenopausal women with osteoporosis.

Results: There were no significant differences in age, body mass index, years since menopause, and initial BMD among the four groups. One-way analysis of variance (ANOVA) with repeated measurements showed a significant decrease in BMD in the C group (P < 0.001). Two-way ANOVA with repeated measurements showed a significant increase in BMD in the D and K groups compared with that in the C group (P < 0.05 and P < 0.001, respectively), and a significant increase in BMD in the DK group compared with that in the C, D, and K groups (P < 0.0001, P < 0.05 and P < 0.01, respectively). These findings indicate that combined administration of vitamin D3 and vitamin K2, compared with calcium administration, appears to be useful in increasing the BMD of the lumbar spine in postmenopausal women with osteoporosis.

In a small study of children with thalassemia, the combination of K2 and calcitriol improved lumbar spine bone density over the course of a year.

Aim of the study: Thalassemic osteopathy (TOSP) has emerged as a topic of interest, as the optimized transfusion regimens and iron chelations has markedly improved the survival of the patients suffering from thalassemia major (TM) and increased the life expectancy.

Results: We also found a decrease in the ratio of undercarboxylated osteocalcin to carboxylated osteocalcin, however, this was not found to be significant. Although the natural course of TOSP is worsening or at least stabilizing, our pilot study demonstrated that vitamin K2 and calcitriol combination clearly has a positive effect on the bone mineral density of the children with TM during a 1-year period. Supplementation of menaquinone-7 instead of drugs is an augmented physiological intake and seems a beneficial alternative for the treatment of TOSP. Further studies on a large number of participants are necessary to highlight the effect of vitamin K2 on TOSP.

In children receiving dialysis, the combination of K2 and vitamin D improved markers related to calcification regulation, although it did not change FGF23.

Aim of the study: Vascular calcification is one of the most prevalent disorders in pediatric hemodialysis patients that eventually lead to cardiovascular morbidity. Vitamin K2 was investigated in adults in previous studies and showed favorable effects on calcification markers. Our aim in this study was to evaluate the efficacy and safety of vitamin K2 and cholecalciferol on the calcification regulators in pediatric patients.

Results: At the end of the study period, serum levels of FGF23, dp-uc-MGP, and uc-OC were measured. It was found that serum levels of dp-uc-MGP, uc-OC, and FGF23 were significantly higher (p < 0.05) in the hemodialysis patients as compared to the healthy normal control. After 4 months, group 3 revealed the most significant decrease in dp-uc-MGP, uc-OC as compared to the other groups. However, there was no change in FGF23. Vitamin K2 and native vitamin D showed a beneficial effect on calcification regulators in pediatric hemodialysis patients.

After spinal surgery, patients taking K2, D3, and calcium had a higher rate of complete fusion after six months than the control group.

Aim of the study: To address the early effects of concurrent treatment with vitamin K2 and vitamin D3 on fusion rates in patients who have undergone spinal surgery. Intervertebral pseudarthrosis has been reported after transforaminal lumbar interbody fusion (TLIF) or posterior lumbar interbody fusion (PLIF), especially in patients with osteopenia or osteoporosis.

Results: Seventy-eight patients were included, and nine patients subsequently discontinued because of 2019-nCoV. At six months postoperatively, complete fusion rates were significantly higher in the VK2+VD3 group than that in the control group (91.18% vs 71.43%, P = 0.036). At six months postoperatively, BMD was increased in the VK2+VD3 group and was higher than that in the control group, although there was no significant difference. At three months postoperatively, a significant increase in procollagen type I amino terminal propeptide (91.81%) and a slight decrease in C-terminal end peptide (8.06%) were observed in the VK2+VD3 group. In both groups, the JOA-BPEQ and VAS scores were significantly improved after spine surgery. Administration of vitamin K2 and vitamin D3 can increase lumbar interbody fusion rates, improve clinical symptoms, promote bone information, and avoid further decline in BMD within six months after TLIF or PLIF.Level of Evidence: 3.

In patients with chronic kidney disease, taking K2 together with D3 was associated with a smaller increase in carotid artery wall thickness than taking D3 alone.

Aim of the study: Observational studies have shown that high dietary intake of vitamin K2 is associated with reduced risk of coronary vascular disease and vascular calcification. We assessed the effect of vitamin K2 substitution on the progression of atherosclerosis and calcification in nondialyzed patients with CKD stages 3-5.

Results: The increase of CCA-IMT was significantly lower in the K+D group compared with the D group: from 0.95 ±0.2 mm to 1.01 ±0.3, P = 0.003 vs from 1.02 ±0.2 mm to 1.16 ±0.3, P = 0.003 (ΔCCA-IMT, 0.06 ±0.08 vs 0.136 ±0.05 mm, P = 0.005, respectively). The increase in CACS was slightly lower in the K+D group than in the D group (ΔCACS, 58.1 ±106.5 AU vs 74.4 ±127.1 AU, P = 0.7). In the K+D group, a significant decrease in the level of dp-ucMGP and total OC was observed. A 270-day course of vitamin K2 administration in patients with CKD stages 3-5 may reduce the progression of atherosclerosis, but does not significantly affect the progression of calcification. Vitamin K2 significantly changes the levels of calcification promoters and inhibitors: dp-ucMGP, OC, and OPG.

In human bone cell cultures, K2 increased mineralization induced by the active form of vitamin D3.

Aim of the study: The effect of vitamin K on mineralization by human periosteal osteoblasts was investigated in the absence and presence of 1alpha, 25 dihydroxyvitamin D3 (1,25(OH)2D3). Vitamin K1 and K2, but not vitamin K3, at 2.5 microM enhanced in vitro mineralization when cells were cultured with vitamin K for 20 days after reaching confluence in vitro.

Results: Warfarin blocked both the 1,25(OH)2D3-induced osteocalcin production and the accumulation in the cell layer, and also blocked the 1, 25(OH)2D3 plus vitamin K2-induced osteocalcin production and the accumulation in the cell layer. The 1,25(OH)2D3-induced mineralization promoted by vitamin K2 was probably due to the enhanced accumulation of osteocalcin induced by vitamin K2 in the cell layer. However, we concluded that the mineralization induced by vitamin K2 alone was due to the accumulation of osteocalcin in bovine serum on the cell layer, since osteocalcin extracted from the cell layer was not identified by specific antiserum against human osteocalcin, which does not cross-react with bovine osteocalcin. These results suggest that the mechanism underlying the mineralization induced by vitamin K2 in the presence of 1,25(OH)2D3 was different from that of vitamin K2 alone, and that osteocalcin plays an important role in mineralization by osteoblasts in vitro.

In sinus bone cell cultures, the combination of D3 and K2 increased osteocalcin production and mineralization.

Aim of the study: Several recent studies have indicated that the paranasal sinus bones undergo pathophysiological changes in patients with chronic sinusitis. We examined the mineralization activity of osteoblasts and the production of osteocalcin and cytokines in cultured human osteoblasts derived from ethmoidal bones treated with vitamin D3, vitamin K2, and warfarin to investigate the metabolic effects of these treatments on paranasal sinus bones.

Results: In the bones treated with vitamin D3 plus vitamin K2, osteocalcin production and the ratio of the mineralization of osteoblasts were increased. Warfarin inhibited the promotive effects of vitamin K2 in the presence of vitamin D3. With regard to TGF-beta production, there was quite a difference in response depending on the isoforms. In conclusion, we have demonstrated that these vitamins and warfarin may be useful in improving bone metabolism in paranasal sinus bones, and may additionally improve the pathogenesis of chronic sinusitis.

In a small retrospective study, a product containing vitamin D, K2, and pollen cytoplasm was associated with improvements in several menopause symptoms and selected bone markers, but this does not prove that the combination of D and K2 alone always provides benefits.

Aim of the study: Background: The menopausal transition may be accompanied by several disabling symptoms which impair quality of life. Many women cannot proceed with and/or refuse hormonal replacement therapy (HRT) and, thus, they rely on non-hormonal remedies to manage climacteric syndrome.

Results: Comparing Group A and Group B, the values of CTX significantly differed after treatment (p = 0.0458). In comparison to the baseline, significant differences emerged within Group A for physical, psychosocial, and vasomotor domains of Menopause-Specific Quality of Life (MenQol) (p < 0.05) whereas no significant difference was found for the sexual domain (p > 0.05). Comparing Group A and Group B, significant differences were observed for all MenQol domains (p < 0.05), except for the sexual domain (p > 0.05) after therapy. Conclusions: Based on our findings, the combination of PCP, vitD, and vitK2 could be used for managing vasomotor and mood disturbances associated with menopause, whilst also offering the added benefit of maintaining bone health.

„Interaction Between Vitamins B and D Supplementation Effects: A study showed that supplementation with vitamin D3 and B vitamins significantly decreased bone turnover in elderly individuals. Specifically, the combination of these vitamins improved plasma levels of 25-hydroxyvitamin D and reduced parathyroid hormone levels, which are critical for bone metabolism (58). Positive Correlation: Another study highlighted a positive association between plasma levels of 25-hydroxyvitamin D and both folate and vitamin B12 in adolescents. This suggests that adequate levels of B vitamins may support the metabolism and effectiveness of vitamin D (59,60).”Interaction Between Vitamins B and D Supplementation Effects: A study showed that supplementation with vitamin D3 and B vitamins significantly decreased bone turnover in elderly individuals.
