St. John’s wort has antidepressant effects partly by inhibiting the reuptake of several neurotransmitters.

„St. John’s wort also has antidepressant effects by inhibiting the reuptake of dopamine, serotonin, gamma-aminobutyric acid (GABA), glutamate, and noradrenaline. By raising the levels of these mediators, St. John’s wort undoubtedly triggers contentment, joy, and euphoria.”Dziurawiec działa przeciwdepresyjnie również poprzez hamowanie wchłaniania zwrotnego dopaminy, serotoniny, kwasu gamma-aminomasłowego GABA, glutaminianu i noradrenaliny. Niewątpliwie podnosząc stężenie tych mediatorów dziurawiec wyzwala zadowolenie, radość i euforię.

St. John’s wort should not be taken at the same time as cardiac glycosides, theophylline, warfarin, or cyclosporines, because it reduces their activity.

„Hypericin is an inhibitor of protein kinase C (tyrosinase) and has an antineoplastic effect. Its wound-healing effects have been scientifically confirmed. St. John’s wort should not be taken at the same time as cardiac glycosides, theophylline, warfarin, or cyclosporines. St. John’s wort reduces their activity.”Hiperycyna jest inhibitorem kinazy białkowej C (tyrozynazy) i daje efekt antyneoplastyczny. Potwierdzono naukowo działanie gojące rany. Dziurawca nie należy zażywać równocześnie z glikozydami naparstnicy, teofiliną, warfarinem i cyklosporynami. Dziurawiec obniża ich aktywność.

Hyperforin stimulates cytochrome P450 enzymes, including CYP3A4 and CYP2C9.

„They improve well-being and help with mental relaxation. This is why St. John’s wort has been used to treat depression. Hyperforin induces the enzymes cytochrome P450, CYP3A4, and CYP2C9 by binding to the Pregnane X Receptor (PXR). A St. John’s wort fraction rich in naphthodianthrones, phloroglucinols, and xanthones has been described as an ingredient in antidepressant preparations.

„Naphthodianthrones, phloroglucinols, and xanthones are monoamine oxidase (MAO) inhibitors. A fraction rich in these compounds is an ingredient in antidepressant preparations. Hypericin increases the skin’s sensitivity to UV rays, which can be used appropriately to treat psoriasis, acne, and vitiligo.”Naftodiantrony, floroglucyny i ksantony są inhibitorami monoaminoksydazy MAO.

St. John’s wort oil has been described as an antidepressant.

„An excellent antidepressant, bile-forming, anti-gallstone, and strengthening remedy. It regulates metabolism and cleanses the body. It improves well-being and supports thought processes. When applied externally, it treats hard-to-heal wounds and rashes. Take 1 teaspoon by mouth in the morning.”Doskonały środek przeciwdepresyjny, żółciotwórczy, przeciwkamiczy i wzmacniający. Reguluje przemianę materii, oczyszcza organizm. Poprawia samopoczucie, usprawnia procesy myślenia. Zastosowany zewnętrznie leczy trudno gojące się rany i wypryski. Zażywać doustnie po 1 łyżeczce rano.

A review indicates that St. John’s wort helps with mild and moderate depression in the short term, but may have significant interactions with medications.

Aim of the study: St. John’s wort is an herb commonly used in Europe for decades and more recently the topic of scientific investigation in this country.

Results: St. John’s wort has been found more effective than placebo and equally as effective as tricyclic antidepressants in the short-term management of mild-to-moderate depression. Comparisons to selective serotonin reuptake inhibitors have provided equivocal data. While it is generally well tolerated in clinical use, there is accumulating evidence of significant interactions with drugs. This evidence-based presentation of the literature includes a brief description of pharmacodynamics and clinical applications, followed by a systematic review of adverse effects, toxicity, and drug interactions.

A review of observational studies suggests that St. John’s wort is well tolerated and may be effective for mild and moderate depression.

Aim of the study: We present a systematic review of observational studies of hypericum extracts in the treatment of depressive disorders. We included non-randomized studies with at least 100 patients suffering from depressive disorders treated with hypericum mono-preparations for at least 4 weeks, which reported clinical outcomes.

Results: Response rates were 60% and 69%, respectively, and the proportions of patients dropping out due to side effects were 3.4% and 5.7%, respectively. Serious side effects or interactions were not reported in any study. The quality of reporting was insufficient in the majority of publications. The available studies show that hypericum extracts are well tolerated and seem to be effective in routine treatment of mild to moderate depressive disorders.

A review reports that St. John’s wort preparations with low hyperforin content outperformed placebo and worked similarly to imipramine and fluoxetine, and that St. John’s wort may affect how other medications work.

Aim of the study: Recently, interactions of herbal medicines with synthetic drugs came into focus of particular interest. In the past 3 years, more than 50 papers were published regarding interactions between St.

Results: Products that do not contain substantial amounts of hyperforin (<1%) have not been shown to produce clinically relevant enzyme induction. On the other hand, some evidence suggests that hyperforin may also contribute to the antidepressant activity of SJW. However, clinical studies using SJW preparations with a low hyperforin amount (<1%) clearly demonstrated the superiority of this plant extract over placebo and its equivalence to imipramine and fluoxetine in the treatment of mild to moderate forms of depression. In the present paper clinical significant SJW interactions are critically evaluated against the background of hyperforin.

A review indicates that St. John’s wort worked better than placebo for mild and moderate depression, although its advantage over standard antidepressants is not conclusive.

Aim of the study: St. John’s wort (SJW) extracts are currently being used to treat depression of various degrees of severity. While many studies have shown it to be superior to placebo, data regarding the effectiveness of using SJW as a stand-alone treatment compared with standard antidepressants has yet to be proven conclusively. This study aims to understand the advantages and disadvantages of SJW as a treatment modality for depression.

Results: In patients with mild and moderate depression, SJW proved superior to placebo. Certain studies comparing the efficacy of SJW versus selective serotonin reuptake inhibitors (SSRIs), especially fluoxetine, reported SJW to be more efficacious, while the majority reported no significant difference. Tricyclic antidepressants were also found to have similar efficacy as SJW. Moreover, treatment with SJW was also found to reduce postmenopausal depression. Regarding the safety profile, although SJW is better tolerated with fewer adverse effects when compared to standardized antidepressants, its predisposition to causing fatal serotonin syndrome, when used in conjunction with other serotonergic agents and drug interactions noted with CYP 450 drugs, raises a question in the safety profile. It is essential to acknowledge that SJW has been used as a treatment measure in Germany.
