In mice with an Alzheimer's disease model, ovocystatin improved memory test scores, especially at the higher dose.
Aim of the study: Cystatin C plays an important role in the course of neurodegenerative diseases and has a beneficial effect by inhibiting cysteine proteases and amyloid-β aggregation. It also induces proliferation and autophagy. Cystatin isolated from chicken egg white, called ovocystatin, has been widely used in medical and pharmaceutical research because of its structural and biological similarities to human cystatin C.
Results: The study results indicate that ovocystatin has a beneficial effect on cognitive functions in APP/PS1 transgenic mice. The strongest effects were found in the group of mice with AD that received ovocystatin in their drinking water at a dose of 40 μg/mouse (p < 0.05). During the Morris water maze trial, mice from the AD group swam significantly farther in the target zone than mice in the AD (vehicle) group (p < 0.05). These results encourage further research into its protective effect, which could be used as an adjunct in treating declining cognitive functions.

In young rats, oral administration of ovocystatin was associated with better memory test results than placebo.
Aim of the study: The positive effect of human cystatin C on the development of Alzheimer's disease has been reported, as it inhibits the formation of β-amyloid oligomers and amyloidogenesis. Cystatin C has been found to have a neuroprotective effect by inhibiting cysteine proteases and inducing autophagy and neurogenesis.
Results: The group of young rats that received ovocystatin orally achieved significantly better results in the MWM than the placebo group (p < 0.05). Similarly, the group of young rats receiving Coloco orally achieved better results in the MWM than both the placebo group and the group of rats receiving Colostrinin (p < 0.05). There were no statistically significant differences between oral and intraperitoneal administration of ovocystatin, Coloco, and Colostrinin in the group of old rats. The results suggest that oral administration of ovocystatin and Coloco has beneficial effects on the cognitive functions of young rats.

In a laboratory study, ovocystatin limited beta-amyloid aggregation and its toxicity to PC12 cells.
Aim of the study: Amyloid β peptides (Aβ) that aggregate in the brain have potential neurotoxic effects and are believed to be a major cause of Alzheimer's disease (AD). Therefore, inhibiting amyloid polypeptide aggregation appears to be a promising approach to the treatment and prevention of this neurodegenerative disease.
Results: Thioflavin-T (ThT) assays, which determine the degree of amyloid peptide aggregation based on fluorescence measurements, circular dichroism spectroscopy (CD), and transmission electron microscopy (TEM) were used to assess how ovocystatin inhibits amyloid fibril formation. Amyloid beta 42 oligomer toxicity was measured using the MTT test. The results showed that ovocystatin has anti-aggregation activity against Aβ42 and inhibits Aβ42 oligomer toxicity in PC12 cells. This work may help in developing potential substances that can prevent or delay beta-amyloid aggregation, one of the main causes of Alzheimer's disease.
