A review reports that beard lichen extracts show activity against some bacteria, especially Gram-positive bacteria, although some microorganisms are resistant to them.

Aim of the study: The main aim of this review is to provide an up-to-date overview of the antimicrobial activities of Usnea sp., including the traditional and medicinal uses, and a critical evaluation of the presented data. Also, the mechanism of this type of action will be explained.

Results: Analysis of the scientific literature regarding traditional uses and bioactivity research showed that Usnea sp. extracts exhibit high antibacterial activity. The Gram-positive bacteria (Staphylococcus aureus, Bacillus subtilis, Bacillus cereus, and Mycobacterium tuberculosis) and aquatic oomycetous fungi were the most sensitive Usnea sp. extracts. Moderate activity against Malassezia furfur and dermatophytes was observed, as well. Gram-negative bacteria, yeast, and fungi were more frequently resistant to Usnea sp. extracts (included Escherichia coli, Candida sp., Saccharomyces cerevisiae, and Aspergillus sp.). The antiviral activity of Usnea sp. was limited. The results show that the use of Usnea sp. in traditional medicine can be scientifically documented. Studies show that usnic acid is the active compound present in Usnea sp. extracts.

In rats infected with staphylococcus, Usnea longissima extract reduced markers of inflammation and oxidative stress in the lungs.

Aim of the study: The role of oxidants and proinflammatory cytokines in the pathogenesis of pneumonia caused by Staphylococcus aureus (S. aureus) has been demonstrated.

Results: S. aureus inoculation caused severe mononuclear cell infiltration in interstitial areas, severe lymphoid hyperplasia in bronchial-associated lymphoid tissue and severe alveolar edema, histopathologically. Treatment with ULEtOAc had an attenuating effect on these histopathological findings. Experimental results from this study suggest that ULEtOAc may be beneficial in treating S. aureus-induced oxidative and inflammatory lung damage.

In cell studies, diffractaic acid from lichens inhibited the multiplication of dengue virus and showed activity against several other viruses.

Aim of the study: Dengue is a mosquito-borne flavivirus that causes 21,000 deaths annually. Depsides and depsidones of lichens have previously been reported to be antimicrobials.

Results: Their EC50 (SI) values were 2.43 ± 0.19 (20.59), 0.91 ± 0.15 (13.33), and 17.42 ± 3.21 (8.95) μM, respectively. Diffractaic acid showed the highest selectivity index, and similar efficacies were also found in dengue serotypes 1-4, Zika, and chikungunya viruses. Cell-based studies revealed that the target was mainly in the late stage with replication and the formation of infectious particles. This report highlights that a lichen-derived diffractaic acid could become a mosquito-borne antiviral lead as its selectivity indices ranged from 8.07 to 20.59 with a proposed target at viral replication.

In a mouse model of infected wounds, a hydrogel dressing containing usnic acid reduced infection and inflammation and supported healing.

Aim of the study: Bacterial cellulose is an extracellular polysaccharide produced by microorganisms, offering advantages such as high water-holding capacity, flexibility, and biocompatibility. However, its lack of bactericidal activity hampers its wide application.

Results: In this study, a hydrogel membrane is designed by combining bacterial cellulose and usnic acid suspended in sanxan gel through a simple in situ microorganism fermentation. The obtained membranes demonstrate excellent ability for sustained drug release, strong eradication capability against tested bacteria in both in vitro and in vivo experiments, effective inhibition of biofilm formation, and excellent hemocompatibility and cytocompatibility. Additionally, the composite membranes promote wound healing with reduced inflammation and bacterial infection in a full-thickness wound infection model in mice. This study provides innovative insights and strategies for the development of functional dressings for infected wounds in future clinical applications.
