A review found that high-quality studies do not link aluminium-adjuvanted vaccines with serious or long-term health problems.
Aim of the study: To systematically review and critically appraise human evidence on potential health effects of aluminium-adjuvanted vaccines.
Results: The review included 59 studies (37 case series, 11 randomised controlled trials, nine cohort studies and two ecological studies). High-quality evidence from randomised controlled trials and large cohorts consistently showed no association between aluminium-adjuvanted vaccines and serious or long-term health outcomes, such as asthma, autism spectrum disorders or other chronic conditions. Studies on macrophagic myofasciitis were generally small and methodologically limited, and did not provide credible evidence of a causal association (very low certainty). Localised, persistent nodules or granulomas were observed infrequently after diphtheria-tetanus-pertussis vaccines, consistent with delayed-type hypersensitivity (<1%, self-limited; moderate to low certainty). For common adverse events (e.g. headache, myalgia), high-certainty randomised controlled trials found no consistent increase in risk with aluminium-adjuvanted formulations.

Aim of the study: Aluminum salts have been used as adjuvants in vaccines for nearly a century, enhancing the immune response to purified antigens and ensuring durable protection against serious infectious diseases. Despite their longstanding record of safety and effectiveness, aluminum adjuvants have become a focus of public concern, with claims linking them to developmental, neurologic, allergic, and autoimmune diseases.
Results: Large-scale clinical and epidemiologic studies consistently demonstrate no association between aluminum-adjuvanted vaccines and autism spectrum disorder, neurotoxicity, allergic disease, or autoimmune disease. Clinically, vaccines adjuvanted solely with aluminum salts generally do not result in systemic reactogenicity, although local reactions are common. Collectively, the evidence strongly supports the safety of aluminum adjuvants and their necessity in certain vaccines. Clinicians can reassure caregivers that aluminum-containing vaccines provide clear benefits, with risks largely limited to transient local reactions and no systemic toxicity signal in large clinical and epidemiologic studies.

The review found no link between aluminium toxicity and vaccines, and a large epidemiological study disproved the hypothesis of a link with MMF syndrome.
Aim of the study: French people have never been so wary about vaccines. The use of aluminum salts in vaccine adjuvants to enhance effectiveness is one of the major reasons for this lack of confidence.
Results: Aluminum levels in biological media of vaccinated subjects are not different from those of unvaccinated subjects. This is consistent with the very small amount of aluminum contained in one dose of vaccine. Indirect toxicity of aluminum was suggested to explain macrophagic myofasciitis in humans in 2011, a disease that could be mediated by an autoimmune/autoinflammatory mechanism. This hypothesis has recently been refuted in a large pharmaco-epidemiological study proving that aluminum-containing adjuvants of vaccines are not responsible for this autoimmune/autoinflammatory syndrome.
